Yup ;) As long as you have the required amount of experience points to unlock the den, you can use it! Incase you aren't aware of how many experience you need, you need around 25,000 points. A brief quote fro the FAQ to back that up:
As you play WolfQuest, complete the missions, hunt, and interact with other wolves, you earn experience points, shown in your player badge (near the top left of your game screen) and in your player stats. As you earn points, new things should become available to you within the game. Here’s what’s programmed:
  • •1500 points- you can name your pups
•6000 points- your ‘pee prowess’ goes up 1 level (each level reduces the number of territory markings required by about 15-20%)
•10,000 points- pee prowess level 2
    •25,000 points- unlock a 4th den choice & pee prowess level 3
•50,000 points- unlock the ability to have a pure white pup & pee prowess level 4
•75,000 points- pee prowess level 5
Source; What are experience points and what do they do?; WolfQuest FAQ - http://www.wolfquest.org/bb/viewtopic.php?f=35&t=34830

And when using the den, remember to report any bugs/glitches you find by using the steps located here (since then we'l be able to get them fixed so users won't have to experience them, particularly our newer players!:

What do I do if I find a bug or glitch?:

Oh, thanks for posting that up! =] I have gotten as far as the white pup, so I should have been able to unlock and gain access to the fourth den. Perhaps it was just a glitch that I didn't entirely think of all that much at the time.

No problem! Also, for the forth den, you need the experience points before you have chosen a den in order to access it, just to let you know ;D

When I make plans on using the Slough Creek den while still finding a mate on Amethyst Mountain, I always seem to forget to rack up the experience points necessary to access it. xD

I also love just playing around with the den in singleplayer, looking for glitch details to include in reports; just earlier this morning I noticed a speck way high in the sky while playing there and, upon using my scent view, found out it was a hovering elk carcass. ^^

I do. =3 I usually try to get as many experience points I can before going on to the second episode, usually by interacting with other wolves or hunting with my mate.

Well, I use it sometimes. I don't think it's a very good den... my pups randomly stray from the den and drown themselves, my mate gets frozen and takes literally four minutes until it gets un-frozen. Other than that, it's a good den..

Yeah I like the SC dens view, but I barely use it because the glitches were so bad that I had to delete the file. If it didn't have so many glitches, it would be my top den to live in. Without these glitches, it would be rated the safest den that there is in SC in my opinion. Some glitches are just soooooooo irritating, and it makes me mad.

I love the den. The view, the nearby water, the nearby cattle ranch. Everything, except the glitches. They're difficult to deal with and I often freeze there.

I like it in Multiplayer, but hate it in Single player. -.- It's so glitchy

Same here. I used it once in single and deleted the game before I even finished marking it. Too glitchy! I use the Bison Peak Cutoff den myself.

I don't use it at all anymore. I used it once, but so many glitches, freezes, and such. I don't go on MP anymore unless I patrol.

I didn't used to do MP, but now I do it more often...
